Procedure

The reagent usually must be prepared before use according to kits of the chemicals
الكاشف ربما يحتاج تحضير قبل الاستخدام حسب النشرة المرفقة مع الكيماويات المستخدمة

--In serum or plasma

-put 1 ml of working reagent in two test tubes
نضع 1 مللى من الكاشف المحضر مسبقا فى انبوبتين اختبار

-put 100 ? of serum in one tube and read after exact 30 sec. abs1 at wave length 500 then wait for additional 2 minutes and read again  abs2
نضع 100 ميكرون من العينة (السيرم) فى احدى الانبوبتين ونبدا بالعد فى عداد الساعة حتى 30 ثانية ونقيس ثم ننتظر دقيقتين اضافيتين ثم ناخذ القرائة الثانية

-put 100 ? of standard in the other tube read after exact 30 sec. abs1 then wait for additional 2 minutes and read again abs2

Calculation

abs of sample (abs2 - abs1) / abs of standard (abs2 - abs1) * 2 = .......mg / dl
فرق القراءات للعينة مقسوم على فرق القراءات للاستاندارد ونضرب فى 2

In urine

the urine must be collected for 24 hrs. then take 1ml urine and put on it 49 ml water then take from the solution 100 ? and repeat as shown above
القياس فى البول يتم عن طريق تجميع البول فى 24 ساعة ثم يحضر 1 مللى بول على 49 مللى ماء ثم ناخذ من التحضير 100 ميكرون ونبدا كما فى السيرم

Calculation

abs of sample / abs of standard * 100 = ....... mg / dl
هنا نضرب فى 100 بدل 2

Normal values

in serum or plasma 0.6 - 1.4 mg / dl
in urine 1  - 1.5 mg / dl
